KPV is a tripeptide, three amino acids in the order lysine, proline, valine, and the smallest molecule in the range: its molecular formula is C16H30N4O4, its molecular weight about 342.4 g/mol and its CAS number 67727-97-3. It is the C-terminal end, residues 11 to 13, of alpha-melanocyte-stimulating hormone (α-MSH), the same parent hormone Melanotan II is modelled on. The full hormone signals pigment cells through a core sequence in its middle. This three-residue tail lacks that core, yet in laboratory work it keeps an action of its own on inflammatory signalling, which is why it is studied as a compound in its own right. It is commonly researched as a separate line of inquiry into the immune and inflammatory signalling of the melanocortin system. The mechanisms under study are its effect on NF-kappaB, a master switch for inflammatory genes, and on the production of inflammatory cytokines. Much of the work is in the gut: a 2008 study reported that it enters intestinal and immune cells through PepT1, a transporter that normally carries small peptides from digested food, and it has been studied in two mouse models of colitis. The evidence is pre-clinical, from cells and animals, and it is not a licensed medicine.
